Description:

O-Parib (Olaparib Tablets) is a potent, oral PARP (poly ADP-ribose polymerase) inhibitor used as targeted therapy for cancers associated with defective DNA repair mechanisms, such as BRCA-mutated ovarian, breast, pancreatic, and prostate cancers. By inhibiting the PARP enzyme, O-Parib prevents cancer cells from repairing their damaged DNA, leading to cell death—especially in tumor cells with existing BRCA mutations.
Olaparib selectively inhibits the PARP enzyme, which is involved in DNA repair. In cancer cells with mutations in BRCA1 or BRCA2 genes, this inhibition leads to the accumulation of DNA damage, resulting in synthetic lethality and selective killing of cancer cells. It spares most normal cells, thereby reducing damage to healthy tissue.

Dosage and Administration
  • Can be taken with or without food
  • Tablets should be swallowed whole, not crushed or chewed
  • Dose adjustments may be required in patients with renal or hepatic impairment
  • Nausea and vomiting
  • Fatigue
  • Anemia
  • Diarrhea or constipation
  • Decreased appetite
  • Headache
  • Dizziness
  • Dysgeusia (altered taste)
  • Perform BRCA mutation testing before initiating therapy
  • Not recommended during pregnancy; may harm the unborn baby. Effective contraception must be used during treatment and for 6 months after the final dose
  • Caution in patients with hepatic or renal impairment
  • Regular blood counts should be monitored due to risk of bone marrow suppression
  • Avoid use in combination with strong or moderate CYP3A inhibitors or inducers
